The Presence and Significance of Bound Aminoazo Dyes in the Livers of Rats Fed p-Dimethylaminoazobenzene*

The general nature of the carcinogenic process induced by chemicals and radiat ion can be conceived of as a release of the cell from normal intrinsic and extrinsic growth controls through (a) an act ivat ion of a latent virus or the formation of a virus, or (b) a heritable change resulting from reactions between the carcinogen and critical physiological entries. Such concepts may correspond to different aspects of the same process. However, regardless of the correct explanation, an objective tha t appears reasonably accessible at the present in the s tudy of the chemical carcinogens is the identification of the compound or set of compounds directly concerned in the init iation of the carcinogenic process. Thus, the parent carcinogen (compound administered) might be converted in the body to a derivative which could be properly considered as the primary carcinogen. Even a part ia l realization of this objective should enable a more logical a t tack to be made on the mechanism of the carcinogenic process. Thus the character of the initial phases in the process might become more evident once the properties of the pr imary carcinogen or carcinogens were known. The identification of the p r imary carcinogen(s) has been the immediate objective of our studies on the hepatic carcinogen, p-dimethylaminoazobenzene. As a working hypothesis, we have considered tha t at least one of the p r imary carcinogens is either p-dimethylaminoazobenzene itself or ~ts metabolite, p-monomethylaminoazobenzene, a :mixture of these dyes, or a compound very closely related to them (14). Our studies on the metabolism of p-dimethylaminoazobenzene (10, 16) and the effect of structure on its ac t iv i ty (13, 15) suppor t this hypothesis.
